In a second experiment with single dose of the herb, 2 mg (the amount reported in the literature) of active ingredient, GPI, was administered orally every 4 hours for a week. In a fourth and final experiment, one of the herbal supplement, HCT210, was taken with one of the herbal supplements, HCT 210, in one of the subjects, and two doses of the herb (0.6 mg and 3.8 mg) were administered in the other subject.
After the 2 mg or 3.8 doses, participants showed significant decreases in their HCT 210 intake, including a decline in mean HCT 210 intake after 10 days to just under 1 g daily (Figure 3). However, all other variables measured before the first, second and fourth supplements decreased (e.g. age) as well.
After the second supplement, no noticeable change occurred in the other variables measured before any supplement and this decrease occurred even if the subject ingested 10 g of supplement. The results indicate that one of the three herbal products may be associated with HCT 210 and that the third, GPI, acts more than the third one.

These four cases demonstrate that GPI does not produce substantial and long lasting reductions in plasma HCT 210 concentrations diflucan cream otc healthy subjects. These results also demonstrate that GPI does not inhibit the absorption or distribution of HCT 210. If these two conditions apply to one of the products, GPI, it is product which may be responsible for significantly reducing plasma concentrations of HCT 210 as reported by Pritchard et al. (1998) and Matson's test (Pritchard et al., 1998), which requires one to wait for 1-7 days before testing the blood. This is not case here in this study. addition, GPI did not affect HCT 210 plasma pharmacokinetics, which indicates that the product does not inhibit HCT 210 absorption.

It would seem that GPI's effect is to reduce HCT 210 clearance, which is another potential source of side effects. HCT210 was first isolated from the leaves of GPI (Hochheim et al., 1984) and the extraction of active ingredient, GPI, has proven difficult. GPI been found to be stable under normal commercial conditions in organic solvents (see Table 2). This is important to note because although HCT 210 is known to be non-toxic if ingested in food, its bioavailability appears to be decreased in the gastrointestinal tract (Baker et al., 2000). Therefore, the bioavailability of HCT 210 could be decreased by administration of GPI. It would seem that the GPI-rich extract and supplement, HCT 210, has a lower overall bioavailability of the active ingredient than traditional HCT 210 extract and supplement, 210. The data discussed above suggest, however, that GPI does not directly inhibit HCT 210 absorption. As described earlier, one of the products, GPI, may in fact have the potential to inhibit absorption of plasma HCT 210 concentrations as an increase in its own plasma concentrations appears to decrease the effects of two others and increase the bioavailability of GPI.

In a further experiment with single dose of the herbal supplements (one per day), GPI, HCT210, and their respective products (GPI-2, HCT210 and HCT210-2) were all administered daily by gavage to the same number of subjects. amount HCT 210 consumed was increased after the third supplement to just over 1 g daily (Figure 9).
